FERMENTATION The core process in biotechnology
Membranes are often installed upstream of the cultivation tank to pre-treat the raw materials. They are used for cleaning, fractionation or concentration during the processing of the products. Furthermore, they are essential for the production of ultrapure water and closed water loops. MICRODYN-NADIR GmbH, which emerged from the membrane activities of Hoechst AG and the AKZO AG, looks back on a wealth of experience in the development, manufacturing and application of membranes and modules. Fermentation is the production of substances facilitated by yeasts, bacteria or cells. These processes are well established in the production of pharmaceuticals and in the food industry. In addition, fermentation processes have gained importance in the so-called “white” or industrial biotechnology for the production of enzymes, bulk chemicals, and, last but not least, biofuels. The isolation of the pure product from the fermentation broth presents a major challenge to modern separation technology. Membrane technology has established itself as a key technology in this field since it offers several unique advantages compared to thermal, physical or chemical separation techniques.

PRODUCTION OF ENZYMES Process scheme
Fermentation and the subsequent treatment with membrane filtration are the core processes in the biotechnology industry. Typical fermentation products include enzymes to be used as additives for foods and washing agents, among others. To achieve high yields in the cultivation tank, the cell concentration in the fermentation broth is kept at a high level. In the process described above the product exists outside the cells. After the broth has left the cultivation tank, suspended solids (cell fragments and bacteria) can be separated from the enzyme solution using a decanter. Subsequently the product solution is treated in a microfiltration unit and then further purification is done in a dia-filtration unit with ultrafiltration (UF) membranes.
During dia-filtration, substances with low molecular weight are washed out of the solution using clean solvent (ultrapure water), while the target molecules are retained by the membrane. SPIRA-CEL® spiral wound modules are recommended because this process requires a large membrane area. The modules consist of PES membranes which are especially suited for this application because of their high flux and closely defined molecular weight cut-off (MWCO). After dia-filtration, the product concentration can be increased by factor 25. Due to the higher concentration of suspended solids, the viscosity of the suspension will increase.

The efficiency of the crossflow filtration of liquids is determined by the correct choice of membrane type and construction of the module as well as the main parameters of the process – pressure and crossflow velocity. The spiral wound module SPIRA-CEL® has the most efficient geometrical design to achieve maximum membrane surface area per volume. Within the spiral wound module, the fluid flows along a spacer net that introduces turbulence. The geometry of the net can be chosen to adjust the entire configuration to meet the needs of the specific process. MICRODYN and SEPRODYN ® capillary and tubular modules are easy to operate because of their open flow channels. The diameters of the capillaries and tubes are chosen according to the demands of the process and depending on the content of solids. The cleaning process is usually facilitated by short back pulses. MICRODYN-NADIR has extensive experience in the evaluation of the influencing factors to optimize membrane processes.

In crossflow filtration the fluid is pumped parallel to the membrane surface. This promotes turbulent mixing during the filtration process and avoids deposition of particles and formation of a fouling layer on the membrane.
