Reagents for Neural Stem Cell Research
B
reakthrough studies have recently rejected the long-standing belief that neuronal tissue is incapable of regeneration. The discovery that neurons, astrocytes and oligodendrocytes arise from neural stem cells located in specific regions of the central nervous system has important clinical applications for the treatment of life-threatening, neuronal diseases, including Parkinson’s Disease. The successful engraftment of neural stem cells following implantation into the brain of rodent models has demonstrated the potential of this cell type. The development of neural stem cell biology requires specific research tools that allow the isolation, expansion and characterization of this stem cell type. In addition, the availability of reagents for the characterization of progenitor cells is critical for the analysis of their therapeutic potential.

Growth Factors Leukemia Inhibitory Factor – An Important Neural Stem Cell Growth Factor
T
he efficacy of CHEMICON’s patented cytokine human Leukemia Inhibitory Factor (hLIF) in the expansion of neural stem cells has recently been reported1. Isolated neural stem cells were cultured in serum-free media containing hLIF, human epidermal growth factor (hEGF) and human basic fibroblast growth factor (hbFGF). Using this system, it was shown that the initial expansion rate of neural stem cells were similar to those cultured in the absence of hLIF (hEGF and hbFGF only). However, neurospheres cultured in the presence of hLIF exhibited superior doubling rates (7-10 days) after 50-60 days in vitro. Significantly, cultures maintained in hLIF consistently produced more neurons than parallel cultures grown in the absence of hLIF. In addition, LIF-treated cells remained multipotent for at least 1 year in vitro, leading to a 107 increase in the cell population. Description

Characterization of Stem Cells and Their Differentiated Progeny
T
he CHEMICON panel of neuroscience reagents includes antibodies specific for neural stem cell markers, including Nestin and Vimentin, and for the phenotypical analysis of differentiated progeny including neurons, oligodendrocytes and astrocytes. This panel also includes reagents for the detection of engrafted human neural stem cells following implantation into immunodeficient mice.
Stem Cell Markers
N
eural stem cells are defined as cells having the ability to self-renew, and to differentiate into multiple phenotypic lineages. Undifferentiated neural stem cells express high levels of the intermediate filament proteins Vimentin and Nestin. Additionally, proliferating neural stem cells usually show little or no expression of mature cell markers.
Using cell sorting technology, it has recently been demonstrated that adult mouse neural stem cells can be purified to 80% homogeneity by using a panel of markers that includes Nestin (high), Heat Stable Antigen (low) and Peanut Agglutinin binding (low)2.

Engraftment Analysis
T
he potential of neural stem cells to be utilized in the treatment of various neuropathies is demonstrated by their successful implantation into neonatal immunodeficient rodents. Engrafted neurons derived from transplanted human neural stem cells can be discriminated from host tissue using antibodies specific for human N-CAM or human nuclei. Proliferating cells of human origin can be detected using an antibody specific for the human nuclear protein Ki-67. This protein is expressed by human cells in all phases of the active cycle and is absent in the resting phase.
Once engrafted, the differentiation of neural stem cells is dependent upon local cues in the host brain. Apart from a few exceptions, neurogenesis is completed soon after fetal development. Consequently, the brain tissue of adult graft recipients lacks the necessary cues needed to drive the differentiation of implanted stem cell progeny. One method to overcome this is the pre-differentiation of neural stem cells prior to implantation. The detection of implanted, pre-differentiated cells can be accomplished by labeling of proliferating and differentiating cells with BrdU prior to implantation. Resulting neurons and glia can be detected using an anti-BrdU antibody.
