RESUMEN

ABSTRACT

El objetivo de este estudio fue determinar la proporción de sexos en crías provenientes de transferencia de embriones e identificar algunos factores que pudieran modificar dicha proporción. Se analizó la información de 2226 becerros, 1222 provenientes de transferencia de embriones (TE) y 1004 de inseminación artificial (IA), obtenidas de 1989 a 1993 en una unidad de transferencia de embriones (CEMEGEN, LICONSA, Estado de México). Para comparar la proporción de sexos de crías nacidas por IA o TE se utilizó la prueba de ji-cuadrada; además, se usó un modelo lineal logarítmico para calcular los efectos de estación, tipo de embrión (fresco o congelado), etapa y calidad del embrión, grado de sincronía donadora-receptora y grupo genético, en la proporción de sexos. Hubo una mayor proporción de machos (p≤0.01) provenientes de TE en comparación con IA (55.4% vs 49.3%). La proporción de machos de TE en verano (61.4%) fue mayor (p≤0.05) que en primavera (51.0%). Las otras variables no modificaron (p>0.05) la proporción de sexos esperada.

The objective of this study was to determine sex ratio in offspring from embryo transfer and to identify factors influencing this proportion. Data of a total of 2226 calves born from 1989 to 1993 at the CEMEGEN LICONSA breeding center, State of México, including 1004 calves born by artificial insemination (AI) and 1222 from embryo transfer (ET) was analyzed. A chi-square test was used to compare sex ratio of calves born by AI or ET. In addition, a logarithmic linear model was used to estimate the effect of season, type of embryo (fresh or frozen), grade and quality of the embryo, donor/recipient synchrony and genetic group on sex ratio. There was a greater proportion of males (p≤0.01) derived from ET (55.4%) than from AI (49.3%). The proportion of males born from embryos collected during the summer (61.4%) was greater (p≤0.05) than in spring (51.0%). The other variables did not modify (p>0.05) the expected sex ratio. INTRODUCCIÓN

t is not easy to obtain replacement heifers of high genetic merit in dairy herds, because nearly all of the females are kept as possible future replacements. The specialized dairy producers would like to control the sex ratio of the offspring born in their herds, seeking mainly females, and thus increase the size of the population to select from (Thibier and Nibart, 1995; Shea, 1999). In both natural mating and artificial insemination (AI), the sex ratio in offspring is 50% male and 50% female (Callesen et al., 1996; Van Wagtendonk-de Leeuw et al., 1998). However, when superovulation and embryo transfer (ET) are used, there is a higher ratio of males, due to factors related to the degree of development and quality of the transferred embryos (Avery et al., 1991; Van Wagtendonk-de Leeuw et al., 1998; Shea, 1999), as well as the sire used in AI (Avery and Quetglas, 1996). The objective of this study was to determine the sex ratio in offspring produced by embryo transfer, and to identify some factors which could modify this proportion.

Los productores lecheros especializados desearían controlar la proporción de sexos de las crías nacidas en sus explotaciones, buscando predominantemente hembras y, de esa manera, incrementar el tamaño de población de la cual seleccionar (Thibier y Nibart, 1995; Shea, 1999). Tanto en monta natural como en inseminación artificial (IA), la proporción de sexos en las crías es 50% machos y 50% hembras (Callesen et al., 1996; Van Wagtendonk-de Leeuw et al.,1998). (TE), hay una mayor proporción de machos atribuida a factores relacionados con el grado de desarrollo y calidad de los embriones transferidos (Avery et al., 1991; Van Wagtendonk-de Leeuw et al., 1998; Shea, 1999), así como del toro utilizado en IA (Avery y Quetglas, 1996). El objetivo de este estudio fue determinar la proporción de sexos en crías provenientes de transferencia de embriones e identificar algunos factores que pudieran modificar dicha proporción.

MATERIALES Y MÉTODOS Este estudio se efectuó con datos de el Centro de Mejoramiento Genético (CEMEGEN) de LICONSA, municipio de Cuautitlán Izcalli, Estado de México, en las coordenadas 19º 43’ N y 94º 14’ O, y 2450 m de altitud. El clima es C(Wo) b(i) en la clasificación de Köppen, que corresponde a templado subhúmedo, con lluvias en verano. La temperatura media anual es 14 ºC, con una oscilación mensual de 5 a 7 ºC; en el invierno hay temperaturas cercanas a 0 ºC y en verano 30 a 35 ºC; la precipitación pluvial promedio es 610.6 mm (García, 1988). Se utilizaron los registros de 2226 crías nacidas en el CEMEGEN, de 1988 a 1993; 1004 crías provenían de TE con información de la donadora y receptora, características de los embriones transferidos y número de crías nacidas, y 1222 de crías de IA. En el CEMEGEN la TE se utiliza para obtener crías de vacas consideradas como élite que posteriormente reemplazarán a las donadoras, mientras que la IA se usa para gestar a las vacas donadoras de embriones, dando un descanso fisiológico mediante la gestación despúes de tres tratamientos superovulatorios. MATERIALS AND METHODS The present study was carried out with data from the Centro de Mejoramiento Genético (CEMEGEN) of LICONSA, municipality of Cuautitlán Izcalli, State of México, 19o 43’ N and 94o 14’ W, at an altitude of 2450 m. The climate is C (Wo) b(i) in the Köppen classification, which corresponds to a sub-humid temperate climate, with rains in summer. The mean annual temperature is 14 oC, with a monthly oscillation of 5 to 7 oC; in winter there are temperatures close to 0 oC, and in summer, 30 to 35 oC. The average rainfall is 610.6 mm (García, 1988). Records of 2226 calves born in the CEMEGEN from 1988 to 1993 were used; 1004 calves resulted from ET with data of the donor and recipient, characteristics of the transferred embryos and number of calves born, and 1222 of IA offspring. In the CEMEGEN, ET is used to obtain offspring from cows considered to be elite, which would later replace the donors, whereas AI is used to gestate the embryo donating cows, thus providing a physiological recess by means of gestation after three superovulatory treatments. Studied variables Sex of the offspring Birth records were obtained at CEMEGEN, indicating whether the origin was AI or ET. Genetic group of the embryo The embryos were obtained from Holstein, Swiss Brown cows, and Bos taurus×Bos indicus (F1).

To compare the sex ratio of the offspring born by AI and ET, the chi-square test was used. A logarithmic linear model was used to calculate the effects of season, type, stage and quality of the embryo, degree of donor-recipient synchrony and genetic group, on the sex ratio (CATMOD; SAS, 1999).

La información fue agrupada por año de estudio, de 1989 a 1993. Análisis estadístico Para comparar la proporción de sexos de las crías nacidas por IA y mediante TE, se utilizó la prueba de ji-cuadrada. Se utilizó un modelo lineal logarítmico para calcular los efectos de estación, tipo, etapa y calidad del embrión, grado de sincronía donadora-receptora y grupo genético, en la proporción de sexos (CATMOD; SAS, 1999).

RESULTADOS Y DISCUSIÓN En el CEMEGEN, de 1989 a1993 nacieron 2226 crías; 54.9% provenía de IA, y 45.1% de TE (Cuadro 1). La proporción de sexos en las crías provenientes de IA, fue 49.3% hembras y 51.6% machos, (p>0.05); para las crías provenientes de TE la proporción no fue 1:1, porque hubo más machos (55.4%) que hembras (44.6%) (p≤0.01). La proporción de machos producida por TE (55.4%) fue mayor (p≤0.01) que la obtenida por IA (49.3%). Esto concuerda con los resultados de Van Wagtendonk-de Leeuw et al. (1998), quienes reportaron 48.9% de machos en los becerros de IA, comparados con 55.5% de machos descendientes de TE producidos in vitro. At CEMEGEN, 2226 calves were born from 1989 to 1993; 54.9% derived from AI, and 45.1% from ET (Table 1). The sex ratio in the AI derived calves was 49.3% female and 51.6% male (p>0.05); for the ET derived calves, the ratio was not 1:1, due to the fact that there were more males (55.4%) than females (44.6%) (p≤0.01). The proportion of males produced by ET (55.4%) was greater (p≤0.01) than that obtained by AI (49.3%). This coincides with the results obtained by Van Wagtendonkde Leeuw et al. (1998), who reported 48.9% males in the AI calves, compared to 55.5% males derived from ET produced in vitro. cuyo promedio fue 55.0%. Según Shea (1999) hay diferencias entre años que se atribuyen a factores aleatorios. Los embriones recolectados en verano (Cuadro 1) produjeron la mayor (p≤0.05) proporción de machos (61.4%), pero esta proporción fue menor en primavera (51.0%), consecuencia posiblemente de mecanismos asociados con el ambiente. Al respecto, factores ambientales como las horas luz, la temperatura y las lluvias afectan los mecanismos reproductivos en algunas especies (Lindsay, 1996; Shea, 1999). En el CEMEGEN, en el verano se registra la temporada de lluvias, ocasionando nublados, encharcamientos en los corrales y otras situaciones adversas para los animales, lo cual, aunado a las altas temperaturas provoca estrés climático. El conjunto de estos factores puede estar ligado con una mayor capacidad de los embriones machos para resistir condiciones adversas y favorecer su mayor proporción; mientras que en las crías de embriones recolectados en la primavera, con un clima más propicio, la proporción de hembras y machos fue 1:1. Shea (1999) observó un efecto estacional, agrupando los meses (p≤0.05) de recolección de los embriones, de acuerdo con una mayor o menor cantidad de horas luz, encontrando una mayor proporción de machos (55.7%) de abril a septiembre cuando hay más horas luz. Según Ealy et al. (1993) las altas temperaturas durante el verano retrasaron el desarrollo embrionario debido a que los embriones tienen una menor capacidad para secretar el interferón σ, que es la señal para el reconocimiento materno de la gestación. Es posible que los embriones hembra, cuyo desarrollo es más lento (Avery et al., 1991), son afectados por las temperaturas del verano y sufrieran un retraso adicional, favoreciendo la proporción de machos.

proportion of males (p≤0.09), with an average of 55.0%. According to Shea (1999), there are differences among years, which are attributed to random factors. The embryos collected in summer (Table 1) produced the greatest (p≤0.05) proportion of males (61.4%), but this proportion was lower in spring (51.0%), possibly due to mechanisms associated with the environment. To this respect, environmental factors such as light, temperature and rain affect the reproductive mechanisms of some species (Lindsay, 1996; Shea, 1999). At the CEMEGEN, the rainy season occurs in summer, causing cloudiness, mud in the lots and other situations which are adverse for the animals, which added to the high temperatures, causes climatic stress. The combination of these factors may be linked to a greater capacity of the male embryos to withstand adverse conditions, thus favoring their higher proportion; whereas in the offspring from embryos collected in the spring, when the climate is more favorable, the proportion of females and males was 1:1. Shea (1999) observed a seasonal effect, grouping the months (p≤0.05) of the embryo collection according to a greater or smaller quantity of light hours, finding a greater proportion of males (55.7%) from April to September, when there are more hours light. According to Ealy et al. (1993), the high temperatures during the summer delayed embryo development, due to the fact that the embryos have a smaller capacity to secrete the interferon σ, which is the signal for maternal recognition of gestation. It is possible that the female embryos, whose development is slower (Avery et al., 1991), are affected by the summer temperatures and suffer an additional delay, thus favoring the proportion of males.

At the CEMEGEN, fresh and frozen embryos were transferred to the recipient cows. There were no differences (p>0.05) in the proportion of male offspring derived from fresh (55.2%) or frozen (54.8%) embryos transferred to the recipient cows. The male embryos have better development (Avery et al., 1991), appearance and quality, and therefore survive manipulation better than the female embryos (King et al., 1992). The combination of these factors may propitiate an artificial selection of the sex of the offspring, due to the fact that the embryos selected to be frozen and transferred tend to be those which are most developed, and thereby with a better appearance. In contrast to the results of the present study, where there were no differences due to the type of embryo, Galli and Lazzari (1996), upon selecting the best embryos for freezing, found a greater proportion of males (60.7%) in the offspring derived from frozen embryos, and only 43.2%

En el CEMEGEN se transferían embriones frescos y congelados a las vacas receptoras. No hubo diferencias (p>0.05) en la proporción de crías machos provenientes de embriones transferidos frescos (55.2%) o congelados (54.8 %) a las vacas receptoras. Los embriones machos tienen un mejor desarrollo (Avery et al., 1991), apariencia y calidad y por tanto sobreviven mejor a la manipulación que los embriones hembras (King et al., 1992). La combinación de estos factores puede propiciar una selección artificial del sexo de la descendencia, debido a que los embriones seleccionados para congelarse y transferirse tienden a ser los embriones más desarrollados y por lo mismo tienen mejor apariencia. En contraste con los resultados del presente estudio, donde no hubo diferencias por el tipo de embrión, Galli y Lazzari (1996), al seleccionar los mejores embriones para

congelamiento, encontraron una mayor proporción de machos (60.7%) en las crías derivadas de embriones congelados y sólo (43.2%) de machos en las crías provenientes de embriones frescos cuya selección no fue muy rigurosa. En el CEMEGEN, para para congelar los embriones se tomaba en cuenta, además de la calidad y grado de desarrollo del embrión, la disponibilidad de receptoras al momento del lavado de las donadoras. Por tanto, se congelaba cualquier embrión transferible, en caso de no haber receptoras disponibles. Este manejo aleatorio del congelamiento de embriones pudo ocasionar que no se encontraran diferencias en la proporción de sexos en las crías de embriones frescos y congelados. Efecto del grupo genético del embrión No se encontraron diferencias (p>0.05) debidas al grupo racial en la proporción de machos: 55.5% de crías machos Holstein, 56.3% Pardo Suizo y 51.9% F1. Efecto de grado de desarrollo y calidad del embrión El grado de desarrollo del embrión no causó diferencias (p>0.05) en la proporción de machos nacidos de embriones transferidos como mórula (58.6%), blastocito temprano (51.5%) o blastocito más desarrollado (54.6%). Seidel (1988) no encontró correlación entre el estado de desarrollo del embrión dentro de la misma donadora y el sexo de las crías resultantes. Sin embargo, en experimentos in vitro se ha documentado una relación significativa entre el sexo del embrión y la tasa de desarrollo (Carvalho et al., 1996; Behboodi et al., 1997; Tocharus et al., 1997; Pegoraro et al., 1998), pero no en todos los estudios (Callesen et al., 1995; Grisart et al., 1995; Roschlau et al., 1997). En experimentos in vitro los embriones machos se desarrollan más rápidamente que los embriones hembras los primeros 8 d después de la fertilización; sin embargo, cuando se determinó el sexo 24 y 40 h después de la fertilización, no hubo diferencias entre machos y hembras, lo que sugiere una pérdida diferencial de embriones hembras durante su cultivo hasta el estado de blastocito (Gutiérrez-Adán et al., 1996). Estos autores encontraron que la proporción de embriones macho/hembra en el día 7 ha llegado a ser 1.97:1 (χ2, p≤0.01); por tanto, se ha sugerido una respuesta diferencial a las condiciones del cultivo in vitro y, específicamente, a los factores de crecimiento u otros componentes del suero que afectan la viabilidad y el desarrollo. Lo anterior afecta la expresión de genes ligados al sexo poco después de la activación del genoma, lo que causa desviación en la proporción de sexos (Wrenzycki et al., 2002).

of males in those from fresh embryos, whose selection was not very rigorous. At the CEMEGEN, in order to freeze the embryos, the availability of recipients at the moment of washing the donors was taken into consideration, as well as the quality and degree of development of the embryos. Therefore, any transferable embryo was frozen, in case no recipients were available. This random handling of the freezing of embryos could explain why no differences were found in the sex ratio of offspring from fresh and frozen embryos. Effect of the genetic group of the embryo No differences were found (p>0.05) due to the racial group in the proportion of males: 55.5% of male offspring of Holstein, 56.3% Swiss Brown and 51.9% F1. Effect of the degree of development and quality of the embryo The degree of embryo development did not cause differences (p>0.05) in the proportion of males born from embryos transferred as morulae (58.6%), early blastocyst (51.5%) or more developed blastocyst (54.6%). Seidel (1988) did not find a correlation between the stage of development of the embryo within the same donor and the sex of the resulting offspring. However, with in vitro experiments, a significant relationship has been documented between the sex of the embryo and the rate of development (Carvalho et al., 1996; Behboodi et al., 1997; Tocharus et al., 1997; Pegoraro et al., 1998), but not in all of the studies (Callesen et al., 1995; Grisart et al., 1995; Roschlau et al., 1997). In experiments in vitro, the male embryos developed faster than the female embryos the first 8 d after fertilization; however, when the sex was determined 24 and 40 h after fertilization, there were no differences between males and females, which suggests a differential loss of female embryos during their culture until the blastocyst stage (Gutiérrez-Adán et al., 1996). These authors found that the proportion of male/female embryos on day 7 had reached 1.97: 1 (χ2, p≤0.01); therefore, it has been suggested that there is a differential response to the in vitro culture conditions, and specifically, to the growth factors or other components of the serum which affect viability and development. The above affects the expression of genes linked to sex shortly after the activation of the genome, causing deviation in the sex ratio (Wrenzycki et al., 2002). The mechanisms which explain the differences in the development of the embryos are not very clear (Pegoraro et al., 1998), but metabolic factors may be involved. These

Los mecanismos que explican las diferencias en el desarrollo de los embriones no son muy claros (Pegoraro et al., 1998), pero pueden estar involucrados factores metabólicos. Estas diferencias dependientes del sexo del embrión sugieren una respuesta diferencial a factores del crecimiento (Gutiérrez-Adán et al., 1996), a otros componentes del suero utilizado para el cultivo de los embriones (Grisart et al., 1995), o la presencia de glucosa (Bredbacka y Bredbacka, 1996). En sistemas in vitro el periodo de la inseminación de los ovocitos hasta la recuperación de los embriones es definido con toda precisión (Dominko y First, 1997). Pero in vivo, la ovulación y la fertilización ocurren en algunas horas; por tanto, las comparaciones directas de resultados en la proporción de sexos obtenidos en los dos sistemas deben hacerse cuidadosamente (Callesen et al., 1995). Debido a que no se evaluaron los diferentes estados de desarrollo del embrión dentro de un mismo lavado, los resultados de este estudio no permiten evaluar lo afirmado por Avery et al., (1991) de no apoyar la hipótesis de un más rápido desarrollo de los embriones machos respecto a los embriones hembras. La calidad del embrión transferido no tuvo efecto (p>0.05) en la proporción de machos nacidos de embriones con calidad excelente (61.0%), buena (55.0%) o pobre (58.0%). Al respecto, Callesen et al. (1995) observaron variaciones de técnico a técnico en la determinación de la calidad de los embriones y, por tanto, no fue posible relacionar la evaluación morfológica de los embriones con el sexo de las crías. Roschlau et al. (1997) tampoco encontraron diferencias en la proporción de sexos en las distintas categorías de calidad de los embriones. Shea (1999) estudió el efecto de la calidad de los embriones en la proporción de sexos, y encontró pequeñas diferencias a favor de los machos (57.0%) en el grupo de embriones de excelente y muy buena calidad, comparados con 53.0% de machos en el grupo de embriones de buena calidad. Sin embargo, se considera insuficiente seleccionar embriones para un determinado sexo utilizando como único criterio la calidad del embrión. Efecto de sincronía La transferencia de embriones con buenos resultados requiere la sincronización entre la edad del embrión y del aparato reproductor de las hembras receptoras. Las pérdidas de embriones por transferencias asincrónicas se deben probablemente a que los embriones se depositan en un ambiente adverso (Krackow, 1997), o a la incapacidad para ejercer acción luteotrópica en el cuerpo lúteo de la receptora. Los efectos de una transferencia asincrónica en la supervivencia de los embriones se manifiestan de manera diferencial en machos y hembras, afectando probablemente más a estas últimas.

differences depending on the sex of the embryo suggest a differential response to growth factors (Gutiérrez-Adán et al., 1996), to other components of the serum used for the embryo culture (Grisart et al., 1995), or the presence of glucose (Bredbacka and Bredbacka, 1996). With in vitro systems, the period of the insemination of oocytes to the recovery of the embryos is precisely defined (Dominko and First, 1997). However, in vivo, the ovulation and fertilization occur within several hours; therefore, the direct comparisons of results in the sex ratio obtained in the two systems should be carefully made (Callesen et al., 1995). Because the different stages of development of the embryo within a same washing were not evaluated, the results of the present study do not permit an evaluation of what was affirmed by Avery et al. (1991), as they do not support the hypothesis of a faster development of the male embryos with respect to the female embryos. The quality of the transferred embryo had no effect (p>0.05) on the proportion of males born of embryos with a quality of excellent (61.0%), good (55.0%) or poor (58.0%). To this respect, Callesen et al. (1995) observed variations from technician to technician in the determination of the quality of the embryos, and therefore, it was not possible to relate the morphological evaluation of the embryos to the sex of the offspring. Neither did Roschlau et al. (1997) find differences in the sex ratio in the different categories of the embryos quality. Shea (1999) studied the effect of quality of the embryos on the sex ratio, and found small differences in favor of the males (57.0%) in the group of embryos with excellent and very good quality, compared with 53.0% of males in the group of embryos of good quality. However, it is considered insufficient to select embryos for a determined sex using the embryo quality as the only criteria. Effect of synchrony In order to obtain good results in embryo transfer, the synchronization of the embryo age and of the reproductive tract of the recipient females is necessary. The loss of embryos due to asynchronic transfers are probably due to the fact that the embryos are deposited into an adverse environment (Krackow, 1997), or to the incapacity to exert luteotropic action in the corpora lutea of the recipient. The effects of an asynchronic transfer on the survival of the embryos are manifested differentially in males and females, probably affecting the females to a greater degree. In the present study, the proportion of male offspring was not different (p>0.05) when the donor-recipient synchrony was −1.0 d (64.5%), 0.5 d (57.5%), 0.0 d (53.0%) or 0.5 d (60.9%), possibly due to the great

En el presente estudio, la proporción de crías machos no fue diferente (p>0.05) cuando la sincronía donadorareceptora fue −1.0 d (64.5%), 0.5 d (57.5%), 0.0 d (53.0%) o 0.5 d (60.9%), posiblemente debido a la gran desproporción en el número de crías entre las clases de grados de sincronía. Finalmente, al analizar los datos para determinar el efecto de sincronía del embrión transferido, respecto de la receptora, no se encontraron efectos (p>0.05) de este factor en la proporción de sexos debido a que en dos grados de sincronía el número de crías nacidas fue muy bajo.

CONCLUSIONES La proporción de crías macho provenientes de transferencia de embriones es mayor que la de hembras; mientras que cuando las crías provienen de inseminación artificial, la proporción de machos y hembras es similar. Entre los factores estudiados en relación con la transferencia de embriones, sólo la época de recolección influyó en la proporción de sexos, observándose mayor cantidad de machos cuando los embriones se recolectaron en el verano, lo cual puede estar relacionado con condiciones ambientales que alteran la fisiología del animal.

disproportion in the number of offspring among the types of degrees of synchrony. Finally, when an analysis was made of the data for determining the effect of synchrony of the transferred embryo with respect to the recipient, no effects of this factor were found (p>0.05) on the sex ratio, due to the fact that the number of offspring born was very low in two degrees of synchrony.

CONCLUSIONS The proportion of male offspring from embryo transfer is greater than that of females; whereas when the offspring are derived from artificial insemination, the proportion of males and females is similar. Among the factors studied in relation to the transfer of embryos, only the season of collection influenced the sex ratio, resulting in a greater quantity of males when the embryos were collected in summer, which could be related to environmental conditions which alter the physiology of the animal. —End of the English version—
