Weekly Maintenance (1) At the Start of the week (Monday) • Clean the reagent containers with distilled or de-ionized water and refill with new reagents. • Make sure to replace the Trypan Blue solution each week. • Clean external surface of capillary tubes, which reach into the sample vial & reagent containers, with a lint-free cloth. • Remove and clean syringe (using 70% ethanol and distilled or de-ionized water).

(Bi-)Monthly Maintenance Perform two Intensive Cleans with 0.1N to 1N HCl instead of Cedex Detergent Procedure • Replace Detergent in reservoir with 0.1N to 1N HCl • Run Prime • Start Intensive Clean • Replace HCl in reservoir with detergent • Run Prime • Run Standard Clean Use Syringe Kit to Backflush flow cell in reverse direction

Syringe Backflush Routine • Detach the tubing and flangeless fittings from position 8 on the multi-way valve (far bottom right hand side of multi-way valve while looking at front of Cedex). • Attach tubing and flangeless fittings from position 8 to receiving port of tubing (supplied in kit). Place open end of tubing into empty beaker. • Unscrew flangeless nut and ferrule from syringe, supplied in kit. • Fill the syringe with DI, particle-free water. • Slide the flangeless nut over frosted tubing leading into waste container (In addition to the frosted tubing, there is also clear tubing leading into the waste container). Do not use this tubing with the nut threads facing the open end of the tubing. • Slip the flangeless ferrule over the tubing, with the tapered position of the ferrule facing toward the nut.

Syringe Backflush Routine • Insert the frosted tubing with the flangeless nut and ferrule in place into the receiving port of the syringe filled with DI, particle-free water and tighten the nut finger tight. • Alternate pushing liquid and air from the syringe through the flowcell by rotating the syringe. Pull the plunger in the reverse direction to allow more air into the syringe. • Continue pushing liquid and air through the syringe. When syringe is empty, detach syringe from tubing and reattach tubing and flangeless fittings finger tight back into position 8. Perform a Standard Clean and check to make sure that there are no leaks coming from position number 8.
