Resumen

The content of insulin-like growth factor 1 (IGF-1) in bovine milk in México is unknown, and high levels could increase the health risk for adult people who ingest milk. The aim of this study was to determine the level of IGF-1 and its relationship with fat content using liquid chromatography with ion trap as a mass detector in milk marketed in Durango, México. During 18 months we analyzed, three times, six lots chosen randomly of whole, reduced-fat, and low-fat milk of three brands. Each sample was pre-treated using cut-off membranes, digested with trypsin, and subjected to liquid chromatography-mass spectrometry. There was a significant difference in the mean concentration of IGF-1 between the brands and types of milk analyzed (Chi square=34.66; p£0.005). The correlation analysis revealed a positive exponential association between IGF-1 concentration and the fat content of milk with determination coefficients of 0.950 for brand 1, 0.974 for brand 2, and 0.984 for recombinant bovine growth hormone-free. To the best of our knowledge, this is the first study documenting an association between IGF-1 levels and fat content in milk. The decision to consume a brand with or without fat depends on the milk taste and IGF-1 requirements.

El contenido del factor de crecimiento insulínico tipo 1 (IGF1) en leche bovina se desconoce y los niveles altos podrían incrementar el riesgo para la salud de los adultos que ingieren leche. El objetivo de este estudio fue determinar el nivel de IGF-1 en leche comercializada en Durango, México, y su relación con el contenido de grasa, mediante cromatografía líquida, con una trampa de iones que actúa como detector de masas. Durante 18 meses se analizaron seis lotes elegidos al azar de leche entera, semi-descremada y descremada de tres marcas, en tres ocasiones. Cada muestra fue pre tratada con membranas que restringen el peso molecular, digerida con tripsina, y se sometió a cromatografía líquida de alta resolución acoplada a espectrometría de masas. Hubo una diferencia significativa en la concentración media de IGF-1 entre las marcas y los tipos de leche analizados (Ji cuadrada = 34.66; p£0.005). El análisis de correlación reveló una asociación exponencial positiva entre la concentración de IGF-1 y el contenido de grasa de la leche, con coeficientes de determinación de 0.950 para la marca 1, 0.974 para la marca 2 y 0.984 para la leche libre de hormona de crecimiento bovino recombinada. Por lo que sabemos, este es el primer estudio que documenta una asociación entre los niveles de IGF-1 y el contenido de grasa en la leche. La decisión de consumir una marca con o sin grasa depende del sabor de la leche y los requerimientos de IGF-1.

Introduction

Introducción

ccording to official estimates, the state of Durango in northern México is the third largest producer of cow’s milk in the country (Lactodata, 2014). In México and other countries, including USA, some dairy producers use recombinant bovine growth hormone (rBGH) to boost cows’ milk production (Machlin, 1973; Daxenberger et al., 1998). However, in Australia, Canada, Japan, and the EU, the sale of rBGH to dairy farmers and its use are prohibited (Cummins, 1999). The use of rBGH stimulates liver production of insulin-like growth factor 1 (IGF-1) (Purup et al., 1993; Epstein, 1996). The IGF-1 is a peptide of 70 amino acids with a molecular weight of 7650 Da (Clemmons, 1997) and its structure is identical in bovines and humans (Honegger and Humbel, 1986), which accounts for the recognition and utilization of the hormone by the human body. The natural concentration of IGF-1 in milk is 1.27-8.10 ng mL-1 (Collier et al., 1991), but IGF-1 concentration in 5777 random milk samples ranges from 1.0 to 83 ng mL-1 (Daxenberger et al., 1998) and 420 ng IGF-1 mL-1 in colostrum (Piot et al., 2004) ranging from 32 to 2,000 ng mL-1 in some cases (Guauthier et al., 2006). Cattle that receive rBGH supplements may suffer mastitis due to an increase of up to 30 % in milk production and udders get swollen, coupled with endocrine effects that cause excessive hormones in the body and the development of diseases such as infertility and problems in the hooves that reduce the cow’s life expectancy due to exhaustion and weakness (Resnicoff et al., 1995). The increment of IGF-1 levels in blood and milk (Daxenberger et al., 1998) and the increased influx of immunoreactive cells from blood into milk, induce a large increase of milk somatic cell counts (Bruckmaier et al., 2004); besides, IGF-1 enhanced mammary cancer progression in rats (Hadsell and Bonnette, 2000; Thordarson et al., 2004). A meta-analysis confirmed a moderately elevated risk of Parkinson disease among people with high dairy consumption (Chen et al., 2007). High quantities of IGF-1 are associated with the appearance of breast, colon, and prostate cancers, due to activity on the pituitary to induce powerful endocrine and metabolic

El IGF1 es un péptido de 70 aminoácidos con un peso molecular de 7650 Da (Clemmons, 1997) y su estructura es idéntica en bovinos y humanos (Honegger y Humbel, 1986), lo que explica el reconocimiento y el uso de la hormona por el cuerpo humano. La concentración natural de IGF-1 en leche es de 1.27 a 8.10 ng mL-1 (Collier et al., 1991), pero la concentración de IGF-1 en 5777 muestras aleatorias de leche es 1.0 a 83 ng mL-1 (Daxenberger et al., 1998) y en el calostro es 420 ng IGF-1 mL-1 (Piot et al., 2004), oscilando de 32 a 2,000 ng mL-1 en algunos casos (Guauthier et al., 2006). El ganado que recibe suplementos de rBGH puede sufrir de mastitis debido al aumento de hasta 30 % en la producción de leche y se hinchan las ubres, junto con efectos endócrinos que causan un exceso de hormonas en el cuerpo y el desarrollo de enfermedades como infertilidad y problemas en las pezuñas que reducen la expectativa de vida de la vaca por agotamiento y debilidad (Resnicoff et al., 1995). El aumento de niveles de IGF-1 en sangre y leche (Daxenberger et al., 1998) y el mayor influjo de células inmunoreactivas de la sangre a la leche inducen un gran incremento en el recuento de células somáticas de la leche (Bruckmaier et al., 2004); y el IGF-1 aumentó la progresión de cáncer de mama en ratas (Hadsell and Bonnette, 2000; Thordarson et al., 2004). Un meta análisis confirmó un riesgo moderadamente elevado de enfermedad de Parkinson entre personas con un alto consumo de lácteos (Chen et al., 2007). Altas cantidades de IGF-1 están asociadas con la incidencia de cáncer de mama, colon y próstata, debido a la actividad sobre la pituitaria al inducir

effects, including cell growth and replication (Ashdown, 1996). Consumption of milk increases IGF-1 concentration in serum levels of infants and it might explain the positive effect in growth (Hoppe et al., 2004); the higher levels of IGF-1 in milk could also favor obesity (Ejlerskov et al., 2014). Rzehak et al. (2013) analyzed 1090 neonates fed milk formula with two protein concentrations (Low = 1.25 and 1.6 g 100 mL milk; High = 2.05 and 3.2 g 100 mL-1 milk) and a control group (588 breastfed children); they found three single nucleotide polymorphisms (SNPs; rs1520220, rs978458 and 2 195 239), that the IGF-1 gene influenced positively the circulating levels IGF1 and the molar concentration of IGF-1 / IGFBP-3 serum, which show that a higher intake of protein metabolic programming contributes to growth. The IGF-1 helps to regenerate damaged muscle mass (Mourkioti et al., 2005), inhibits apoptosis (Dunn et al., 1997), and enhances stem cell recruitment to injured skeletal muscle (Schulze and Spate, 2005). Aging appears to be associated with lower amounts of the growth hormone (GH) and IGF-1, and low calorie intake (Anderson et al., 2009; Bartke, 2011). Milk consumption is associated with high levels of plasma IGF-1 (Morimoto et al., 2005; Rich-Edwards et al., 2007) and fetal weight increment with maternal milk (Heppe et al., 2011); besides, the levels of IGF1 and insulin are diminished if milk is substituted with cola beberages (Hoppe et al., 2009), confirming that milk intake increases IGF-1 levels. Gauthier et al. (2006) used radioimmunoassay (RIA) and report that the IGF-1 content ranged from 0.6 to 150 ng mL-1 for milk and 32 to 2000 ng mL-1 for colostrum. The RIA test is not allowed in our research facilities, because a government permit is needed to handle radioactive substances (DOF, 2012). IGF-1 is also analyzed through an enzymelinked immunoabsorbent assay (ELISA) of serum (Ibrahim et al., 2013). For milk, Castigliego et al. (2011), and Ollikainen and Muuronen (2013) used ELISA, due to its simplicity and reliability provided by a sample pre-treatment. Analysis of IGF-1 can also be performed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF) (Sparbier et al., 2005), and by MALDI-TOF with confirmation of metabolites by Western blotting (King et al., 2009). Hence, we developed an analytical approach using liquid chromatography and mass spectrometry (LC-

El consumo de leche aumenta la concentración de IGF-1 en los niveles del suero de infantes y puede explicar el efecto positivo en el crecimiento (Hoppe et al., 2004); los niveles mayores de IGF-1 en la leche también pueden producir obesidad (Ejlerskov et al., 2014). Rzehak et al. (2013) analizaron 1090 neonatos que fueron alimentados con fórmula con dos concentraciones de proteína (Baja=1.25 y 1.6 g 100 mL-1 leche; Alta = 2.05 y 3.2 g 100 mL-1 leche) y un grupo testigo (588 niños alimentados con leche materna); encontraron tres polimorfismos de nucleótido simple (SNPs; rs1520220, rs978458 y 2 195 239), que el gen IGF-1 influyó positivamente los niveles de circulación de IGF-1 y una concentración molar de IGF-1 / IGFBP-3 suero, lo cual muestra que un consumo mayor de programación metabólica de las proteínas contribuye al crecimiento. El IGF-1 ayuda a regenerar masa muscular dañada (Mourkioti et al., 2005), inhibe la apoptosis (Dunn et al., 1997), y estimula el reclutamiento de células madre en músculo esquelético dañado (Schulze y Spate, 2005). El envejecimiento parece estar asociado con menores cantidades de la hormona de crecimiento (GH) e IGF-1, e ingesta menor de calorías (Anderson et al., 2009; Bartke, 2011). El consumo de leche está asociado con niveles altos de IGF-1 en plasma (Morimoto et al., 2005; Rich-Edwards et al., 2007), y con un aumento del peso fetal con leche maternal (Heppe et al., 2011); además, los niveles de IGF-1 e insulina disminuyen si la leche se sustituye con bebidas de cola (Hoppe et al., 2009), lo cual confirma que el consumo de leche aumenta los niveles de IGF-1. Gauthier et al. (2006) usaron radioinmunoensayo (RIA) y reportan que el contenido de IGF-1 fue de 0.6 a 150 ng mL-1 para leche y 32 a 2000 ng mL-1 para calostro. La prueba de RIA no es permitida en nuestras instalaciones de investigación, porque se requiere un permiso del gobierno para manejar sustancias radioactivas (DOF, 2012). El IGF-1 también se analiza con inmunoabsorción ligada a enzimas (ELISA) del suero (Ibrahim et al., 2013). Para leche, Castigliego et al. (2011) y Ollikainen y Muuronen (2013) usaron ELISA por su simplicidad y confiabilidad proporcionada por un pretratamiento de la muestra. El análisis de IGF-1 también se puede realizar por espectrometría de masas con desorción/ ionización láser asistida por matriz (MALDI-TOF)

MS), with a cut-off pre-treatment of the sample, to quantify IGF-1 levels in milk commercially available in Durango, northern México. The pre-treatment method was based on that used by Bobe et al. (1998) and Ollikainen and Muuronen (2013) with LC-MS, to determine the concentration of IGF-1 in whole, reduced-fat, and low-fat milk of three brands sold in Durango. Milk fat was analyzed using the 969.16 method (AOAC, 1997). So far, a safe level of IGF1 in milk has not been established; in childhood it is associated with healthy growth and helps muscle regeneration, but it is not known whether it is healthy for adults to consume due to potential health risks associated with milk ingestion.

Materials and Methods Chemicals Acetonitrile, formic acid, trypsin type I (10 000 units mg protein-1) from bovine pancreas, IGF-1, guanidine hydrochloride (GdnHCl), bis-tris, dithiothreitol (DTT), sodium citrate, hydrochloric acid (HCl) and Amicon™ ultra centrifugal filter units, were purchased from Sigma-Aldrich® (St. Louis, MO, USA). Milk sampling Three brands of cow’s milk marketed in Durango were used to carry out the study from September 2011 to February 2013. All the samples were packed in Tetra Pak™ and taken randomly from the main grocery stores. Two out of the three brands cover more than 80 % of the milk consumed in Durango (Brands 1 and 2); the third brand was chosen as a control, labeled as rBGH-free, because the producers feed their cows with their own products free of pesticides and herbicides and they do not inject cows with hormones. In México, it is not forbidden to inject cattle with rBGH. For each brand, whole, reduced-fat, and low-fat milk were tested. The samples were replicated six times (3×3×6=54 samples), as recommended by the power test (Montgomery, 2002), taking the average of three analysis from each sample. Chemical analysis Besides the analysis of milk fat (method 969.16), the milk parameters required by the Mexican Norm (NOM, 2003) were analyzed: water (method 948.1), proteins (method 991.23) and lactose (930.28), which were determined according to AOAC,

(Sparbier et al., 2005), y por MALDI-TOF con confirmación de metabolitos por electrotransferencia (Western blot) (King et al., 2009). Así desarrollamos un enfoque analítico con cromatografía líquida y espectrometría de masas (LC-MS) y un pretratamiento de filtros de restricción de peso molecular de la muestra, para cuantificar los niveles de IGF-1 en leche disponible comercialmente en Durango, en el norte de México. El método de pretratamiento se basó en uno usado por Bobe et al. (1998) y Ollikainen y Muuronen (2013) con LC-MS, para determinar la concentración de IGF-1 en leche entera, semi-descremada y descremada de tres marcas vendidas en Durango. La grasa de la leche se analizó con el método 969.16 (AOAC, 1997). Hasta ahora no se ha establecido un nivel seguro de IGF-1 en la leche; en la niñez está asociado con un crecimiento saludable y ayuda a la regeneración muscular, pero se desconoce si afecta a los adultos que lo consumen debido a riesgos potenciales de salud asociados con la ingesta de leche.

Materiales y Métodos Químicos En Sigma-Aldrich® (St. Louis, MO, USA) se compró acetonitrilo, ácido fórmico, tripsina tipo I (10 000 unidades mg proteína-1) de páncreas de bovino, IGF-1, hidrocloruro de guanidina (GdnHCl), bis-tris, ditiotreitol (DTT), citrato de sodio, ácido hidroclórico (HCl) y unidades de filtración para ultra centrífuga marca Amicon™. Muestreo de leche Tres marcas de leche de vaca comercializada en Durango se usaron para realizar el estudio de septiembre 2011 a febrero 2013. Todas las muestras estaban empacadas en Tetra Pak™ y se eligieron al azar de tiendas de abarrotes principales. Dos de las tres marcas cubren más de 80 % de la leche consumida en Durango (Marcas 1 y 2); la tercera marca se eligió como testigo, etiquetada como libre de rBGH, porque los productores alimentan sus vacas con sus propios productos libres de pesticidas y herbicidas y no las inyectan con hormonas. En México, no está prohibido inyectar al ganado. Para cada marca se analizó leche entera, semi-descremada y descremada. Las muestras se replicaron seis veces (3×3×6=54 muestras), como lo recomienda la prueba de poder (Montgomery, 2002), tomando el promedio de tres análisis de cada muestra.

(1997), in order to determine whether the national specifications of milk are fulfilled (Table 1). Pre-treatment of the samples Two mL of milk were dissolved in 2 mL of a pH 6.8 buffer composed of 6 M GndHCl, 0.1 M bis-tris, 19.5 mM DTT, and 5.37 mM sodium citrate (Bobe et al., 1998). The buffer was used to allowing protein denaturation, breaking the sulfursulfur bonds and decoupling them from the binding receptors. The mixture was transferred to a 10 kDa Amicon™ cut-off filter, centrifuged at 4000 × g for 30 min, and rinsed with 2 mL of buffer. The retentate, containing as1-casein, as2-casein, b-casein, k-casein, a-lactoglobulin and b-lactoglobulin, because of their molecular weights higher than 10 000, was discarded. The flowthrough or eluate was then transferred to a 3-kDa Amicon™ cutoff filter, centrifuged at 20 000 × g for 20 min, and rinsed with 2 mL of the buffer to eliminate lactose, salts, and low molecular weight components. The Amicon™ filter was inverted to retrieve the retained solution, which was reconstituted with 200 mL of 1 mg mL-1 trypsin solution in 1 mM HCl, and digested for 18 h at 37 °C. The digested sample was filtered at 20 000 × g through a 3000 Mw Amicon™ cut-off filter, rinsed with a solution of acetonitrile:water:formic acid (100:900:1) to a pH of 4.3, and brought to a final volume of 200 mL (10 times preconcentrated). The retentate was discarded. LC-MS analysis A Varian 500-MS LC™ ion trap mass spectrometer with electrospray ionization (ESI), (Agilent Technologies, México), was used to perform the analysis of IGF-1. Firstly, the conditions were optimized to generate fragments from intact IGF-1, including needle voltage, spray shield, nebulizer gas (nitrogen), drying gas pressure rate (air), and drying gas temperature. Once the parameters were optimized, 20 mL of each individually digested sample were injected using a Rehodyne™ 6 port

Análisis químico Además del análisis de la grasa de leche (método 969.16), se analizaron los parámetros requeridos por la Norma Mexicana (NOM, 2003): agua (método 948.1), proteínas (método 991.23) y lactosa (930.28), los cuales fueron determinados con AOAC (1997), para establecer si las especificaciones nacionales de leche se cumplen (Cuadro 1). Pretratamiento de las muestras Dos mL de leche se disolvieron en 2 mL de un amortiguador pH 6.8 compuesto por 6 M GndHCl, 0.1 M bis-tris, 19.5 mM DTT y 5.37 mM citrato de sodio (Bobe et al., 1998). El amortiguador se usó para permitir la desnaturalización de la proteína, al romper los enlaces azufre-azufre, y los separa de los receptores vinculantes. La mezcla se transfirió a un filtro de corte de 10 kDa Amicon™, se centrifugó a 4,000 × g por 30 min, y se enjuagó con 2 mL de amortiguador. El retentado, que contenía as1-caseína, as2-caseína, b-caseína, k-caseína, a-lactoglobulina y b-lactoglobulina, se desechó, debido a sus pesos moleculares mayores de 10 000. El material eluido se transfirió a un filtro de corte de 3-kDa Amicon™, se centrifugó a 20 000 × g por 20 min, y se lavó con 2 mL del amortiguador para eliminar lactosa, sales y componentes de bajo peso molecular. El filtro Amicon™ se invirtió para recuperar la solución retenida, la cual se reconstituyó con 200 mL de 1 mg mL-1 solución de tripsina en 1 mM HCl, y se digirió 18 h a 37 °C. La muestra digerida se filtró a 20 000 × g a través de un filtro de corte de 3,000 Mw Amicon™, se enjuagó con una solución de acetonitrilo:agua:ácido fórmico (100:900:1) hasta un pH de 4.3, y se llevó a un volumen final de 200 mL (10 veces pre-concentrado). El retentado se desechó. Análisis LC-MS Para realizar el análisis de IGF-1, se usó un espectrofotómetro de masa con trampa de iones e ionización de electrospray (ESI) Varian 500-MS LC™ (Agilent Technologies, México). Primero, las condiciones se optimizaron para generar fragmentos desde IGF-1 intacto, incluyendo voltaje de aguja, protector de rocío, nebulizador de gas (nitrógeno), tasa de presión de gas secante (aire), y temperatura de gas secante. Una vez optimizados los parámetros, se inyectaron 20 mL de cada muestra individualmente digerida, usando una válvula manual de inyección de puerto Rehodyne™ (Agilent Technologies, México). La tasa de flujo fue 0.05 mL min-1 con una fase móvil de 70 % de solvent A y 30 % de solvente; el solvente A fue una mezcla 100:900:1 de acetonitrilo:agua:ácido fórmico y el solvente B fue una mezcla 900:100:1 de acetonitrilo:agua:ácido fórmico. injection manual valve (Agilent Technologies, México). A flow rate of 0.05 mL min-1 with a mobile phase of 70 % of solvent A and 30 % of solvent B, were used; solvent A was a 100:900:1 mixture of acetonitrile:water:formic acid and solvent B was a 900:100:1 mixture of acetonitrile:water:formic acid. The separation was performed using a Poroshell 300SB-C18 column (2.1×75 mm; 5 mm particle size) (Agilent Technologies, México). Statistical analysis A total of 54 samples were analyzed during 18 months and for the statistical analysis the Kruskal-Wallis test with the Statistica software version 7.0 was used because the data were not normalized. A calibration curve was constructed plotting each standard of known concentration versus the area of each standard. Linear regression was considered if the Pearson correlation coefficient (r) was > 0.99. The fat content of each type of milk for the three brands was plotted vs. IGF-1 content, and the curve that had the best fit according to the r2 coefficients of determination was found.

Results and Discussion The sequences of the peptide fragments of IGF1 and those resulting from digesting IGF-1 with trypsin were retrieved and blasted from the webpage of the Universal Protein Resource3 (Table 2), and they were organized by position of the fragment. Only fragment 5 had a molecular weight above 3000 Da, and it was retained in the 3000 Da cut-off filter. The ESI chamber and standard optimized parameters were: needle voltage of 5000 V; spray shield of 600 V; nebulizer gas (nitrogen) pressure rate of 241 kPa; drying gas pressure rate (air) of 69 kPa; and drying gas temperature of 350 °C. The peptide fragments at positions 6 (GFYFNKPTGYGSSSR), 8 (APQTGIVDECCFR), and 19 (GSAGNK) gave the highest signals during optimization of the LCMS system for undigested IGF-1. Optimization parameters such as capillary voltage, radiofrequency, and excitation voltage for the highest signals of the most abundant peptides found by LC-MS are summarized in Table 3. The optimal capillary voltage ranged from 150 to 250 V, the frequency from 118 to 183 Hz, and the excitation voltage from 0.94 to 2.55 V. 3

Análisis estadístico Un total de 54 muestras se analizaron durante 18 meses y para el análisis estadísticos se usó la prueba de Kruskal-Wallis con el programa Statistica versión 7.0 porque los datos no estaban normalizados. Una curva de calibración se construyó al graficar cada estándar de concentración conocida contra el área de cada estándar. La regresión lineal se realizó cuando el coeficiente de correlación Pearson (r) era > 0.99. El contenido de grasa de cada tipo de leche de las tres marcas se graficó contra el contenido de IGF-1, y se encontró la curva con el mejor ajuste con base en los coeficientes de determinación r2.

Resultados y Discusión Las secuencias de los fragmentos de péptidos de IGF-1 y los que resultan de la digestión de IGF-1 con tripsina se recuperaron y se descargaron de la página Web de Universal Protein Resource3 (Cuadro 2), y fueron organizados por posición del fragmento. Sólo el fragmento 5 tuvo un peso molecular mayor a 3000 Da, y se retuvo en el filtro de corte 3000 Da. La cámara ESI y los parámetros estándar optimizados fueron: voltaje de aguja de 5,000 V; protección de espray de 600 V; tasa de presión del gas nebulizador (nitrógeno) de 241 kPa; tasa de presión del gas secante (aire) de 69 kPa; y temperatura del gas secante de 350 °C. Los fragmentos de péptidos en las posiciones 6 (GFYFNKPTGYGSSSR), 8 (APQTGIVDECCFR), y 19 (GSAGNK) dieron las señales más altas durante la optimización del sistema LC-MS para IGF-1 sin digerir. Los parámetros de optimización como el voltaje capilar, la radiofrecuencia y el voltaje de excitación para las señales más altas de los péptidos más abundantes encontrados con LC-MS, se resumen en el Cuadro 3. El voltaje capilar óptimo tuvo un rango de 150 a 250 V, la frecuencia de 118 a 183 Hz, y el voltaje de excitación de 0.94 a 2.55 V. mostró que ambos coeficientes de correlación de Pearson fueron > 0.99. La recuperación de IGF-1 de muestras contaminadas de leche con concentraciones altas y bajas de IGF-1 fue de 82 a 86 %. Con base en la tasa de señal de ruido de 3:1, el límite de detección del fragmento 6 fue 0.2 ng mL-1 de leche (Cuadro 4). Las concentraciones de IGF-1 para leche libre de rBGH y semi-descremada se dejaron con fines visuales, aunque ambas fueron menores al límite de cuantificación. La leche entera tuvo el contenido más alto de IGF-1, con 79.7±23.1, 38.8±10.4 y 5.9±2.0 ng mL-1 de leche para la Marca 1, la Marca 2 y la libre de rBGH, respectivamente. Estas concentraciones están dentro del intervalo reportado por Daxenberger et al. (1998), de 1 a 83 ng mL-1 de leche, a partir del análisis RIA de 5777 muestras de leche natural, y dentro del rango de 0.6 a 150 ng mL-1 de leche descrito por Guauthier et al. (2006). Pero son menores a los valores reportados por Ollikainen y Muuronen (2013), quienes usaron análisis pre-tratamiento y ELISA. Las concentraciones de IGF-1 en leche son similares a las reportadas por Daxenberger et al. (1998) y Guauthier et al. (2006), lo cual confirma que usar el amortiguador de pretratamiento reportado por Bobe et al. (1998) seguido de la filtración de corte, genera datos confiables. El análisis con LC-MS usando el fragmento de 1667.8 Mw tiene un tiempo de análisis más corto que el método ELISA, pero LC-MS es menos preciso que ELISA (De Dios et al., 2013). El IGF-1 en la leche entera de la marca libre de rBGH tuvo una baja concentración (5.9±2.1 ng mL-1) y dentro del rango reportado por Colliers et al. (1991) de 1.27 a 8.10 ng mL-1, confirmando que la leche etiquetada como libre de rBGH tuvo cantidades menores de IGF-1. Las altas desviaciones estándares de las muestras sugieren un bajo control de calidad por los productores. La leche entera de cada marca tuvo la concentración de IGF-1 más alta, seguida de la leche semi descremada y la descremada. Los contenidos de proteína fueron 32.7±0.6, 34.4±0.5, y 33.8±0.5 g L-1 de leche para la leche entera, semi-descremada y descremada, respectivamente; todas cumplen con el límite (>30 g L-1 de leche) establecido por la Norma Mexicana. Los contenidos de grasa fueron 36.8±1.2, 18.3±1.5, y 5.5±1.0 g L-1 de leche, para leche entera, semi descremada y descremada, respectivamente. indicated that both Pearson correlation coefficients were >0.99. The IGF-1 recovery of spiked milk samples of low and high IGF-1 concentrations was between 82 and 86 %. Based on a signal to noise ratio of 3:1, the detection limit of fragment 6 was 0.2 ng mL-1 of milk (Table 4). The concentrations of IGF-1 for rBGH-free and low-fat milk were left for visual purposes, even though they were below the limit of quantification. Whole milk had the highest IGF-1 content, with 79.7±23.1, 38.8±10.4 and 5.9±2.0 ng mL-1 of milk for Brand 1, Brand 2, and rBGH-free, respectively. These concentrations are within the range reported by Daxenberger et al. (1998), of 1 to 83 ng mL-1 of milk, from the RIA analysis of 5777 samples of natural milk, and within the range of 0.6 to 150 ng mL-1 of milk described by Guauthier et al. (2006). However, they are lower than the values reported by Ollikainen and Muuronen (2013), who used pre-treatment and ELISA analysis. The IGF-1 concentrations in milk are similar to those reported by Daxenberger et al. (1998) and Guauthier et al. (2006), which confirms that using the pre-treatment buffer reported by Bobe et al. (1998) followed by cut-off filtration generates reliable data. Analysis with LC-MS using the fragment of 1667.8 Mw has a shorter analysis time than the ELISA method, but LC-MS is less accurate than ELISA (De Dios et al., 2013). The IGF-1 in the whole-milk rBGH-free brand had a low concentration (5.9±2.1 ng mL-1) and within the range reported by Colliers et al. (1991) of 1.27 to 8.10 ng mL-1, confirming that milk labeled as rBGH-free had lower amounts of IGF-1. The high standard deviations of the samples suggest poor quality control by milk producers. Whole milk for each brand had the highest concentration of IGF1, followed by reduced-fat and low-fat milk. Protein contents were 32.7±0.6, 34.4±0.5, and 33.8±0.5 g L-1 of milk for whole, reduced-fat, and low-fat milk, respectively; they all fulfill the limit (>30 g L-1 of milk) established by the Mexican Norm.
